Fluorescently labelled molecule delivery

The LumiPore® platform uses pulsed light to generate transient, gentle disruptions in the plasma membrane, allowing extracellular fluorescent cargo to diffuse directly into the cytosol. This enables direct, reproducible cytosolic access for cargo tracking, live-cell imaging, high throughput cell staining and spacial transfection.

Fluorescently labeled molecules are widely used to study cellular processes, intracellular trafficking, molecular interactions, and delivery efficiency. A range of fluorescent cargoes, including fluorophore-conjugated antibodies, nanobodies, dextrans, and proteins, as well as unconjugated fluorophores such as Alexa Fluor dyes, are routinely used to visualize, track, and quantify biological events in living cells. However, most of these molecules exceed the size or polarity limits for passive diffusion across the plasma membrane, requiring an intracellular delivery method to reach the cytosol or other target compartments.

Conventional delivery approaches typically rely on endocytosis, resulting in low cytosolic bioavailability and cell-to-cell variability in intracellular distribution. Fixed-cell staining protocols avoid this problem but require chemical fixation and permeabilization, which halts cellular activity and precludes observation of dynamic processes in real time.

The LumiPore® Solution

The LumiPore® platform addresses these limitations through photoporation: pulsed light is used to generate transient, gentle disruptions in the plasma membrane, allowing extracellular fluorescent cargo to diffuse directly into the cytosol while maintaining cell viability and downstream functionality. By enabling direct, reproducible cytosolic access the LumiPore intracellular delivery platform supports applications including cargo tracking, live-cell imaging, high throughput cell staining and spacial transfection.
